Jayne tries to turn on the doctor & his sister (who are fugitives) while they're in a hospital. The doctor, Simon, has taken her to the hospital to try to figure out what the gov't (or "Blue Sun" or some organization that Joss Whedon never fully figured out or hadn't yet revealed).

Captain Malcolm Reynolds figured it out & almost kicks Jayne (literally) off the ship. Mal to Jayne: "Next time you stab me in the back, have the courage to do it to my face."

As opposed to the movie, Serenity.

At the end of the episode, as Mal has taken on Dr. Simon Tam, his troubled sister (River), and Shepherd Book (who's been out of the monastery two days). Mal offers Simon the job of medic.

Simon: "I'm trying to put this as delicately as I can.... How do I know you won't kill me in my sleep?"

Mal: "You don't know me, son. So let me explain this to you once: if I ever kill you, you'll be awake, you'll be facing me, and you'll be armed."

Simon: "Are you always this sentimental?"

I'm watching these again, so expect more delightful quotes from me.
